Keratin contaminations
Afin de maximiser les chances de succès des analyses de protéines en spectrométrie de masse, il est vivement conseillé de préparer les échantillons selon les recommandations ci-dessous.

- Wearing a clean coat and gloves is essential throughout the manipulation.
- Gloves must be renewed for each manipulation.
- Avoid wearing sweaters made of wool, mohair, etc.
The material: Keratins come from dust in suspension.
- Equipment and dishes that come into contact with the gel should be washed with detergent, rinsed with ultrapure water and dried with ethanol.
- All equipment should be stored away from dust.
- If possible, handle under a laminar flow hood.
The solutions: The solutions themselves can be a source of keratins.
- Preferably use 1.5 ml transparent polypropylene microtubes.
- Use new products that will be exclusively dedicated to gels.
- The cones will be reserved for proteomic use. Wearing gloves!
- Preferably use pre-filled tip boxes to avoid any manipulation of the pipette tips.

- If possible, use thin gels (0.75mm).
- For 1D gels, load the empty wells with Laemmli buffer to limit the diffusion of proteins on the sides during migration.
- Conservation of the gel in 1% acetic acid at 4°C.
- Prefer Coomassie blue staining to silver nitrate staining.
- In the event of staining with silver nitrate, process the samples as soon as possible.
